Oncology Mutation Analysis

MutectorTM Reagents for Oncology

Epidermal growth factor receptor (EGFR) is a member of the ErbB family of receptor tyrosine kinases which also includes ErbB2 (HER2), ErbB3 (HER3) and ErbB4 (HER4).EGFR and its complex signaling pathways play a central role in the regulation of cell proliferation and death. EGFR and its signaling components are promising targets for effective therapy for various cancers. Somatic mutations of EGFR and its downstream pathway components have been shown in a number of different cancers including melanoma, colorectal cancer, non-small cell lung cancer, papillary thyroid cancer, and ovarian cancer.

For example, approximately 35% to 45% of metastatic colorectal cancer (mCRC) tumors may have a KRAS or BRAF mutation, which negatively affects anti-EGFR therapies. On the other hand, approximately 50% of melanoma tumors may have a BRAF mutation, which increases the sensitivity to BRAF and MEK inhibitors. Thus, the detection of mutations in EGFR and its downstream pathway components is greatly important to personalized medicine and proper oncology therapies. TrimGen has developed and validated MutectorTM reagents to detect clinically relevant mutations of BRAF, KRAS, NRAS, and PIK3CA gene.

Pharmacogenetics Testing

The standard dose of a prescribed drug can cause a serious adverse response in one patient and less of a response in another. Genetic variability is directly linked to an individual's drug response. By analyzing the individual variations in the genes that produce enzymes involved in metabolizing drugs, a physician can provide a more effective treatment plan, avoiding adverse drug reaction, toxicity, or lack of drug efficacy.

Cytochrome P450 (CYP) enzymes account for 70-80% of enzymatic activity involved in drug metabolism. The genetic variations of CYP genes affect enzyme activity, which in turn affects drug metabolism. For example, CYP2D6, a member of the CYP oxidase system, was shown to be responsible for the metabolism and elimination of approximately 25% of clinically used drugs. The genetic variations (polymorphism) of the CYP2D6 genes affect its function in drug metabolisms. Thus, the identification of genetic variations of CYP and its downstream pathway components are greatly important to personalized medicine and proper treatment. Based on the ability to metabolize its substrates, an individual’s CYP2D6 function can be categorized as one of the following: poor metabolizer (PM), intermediate metabolizer (IM), extensive metabolizer (EM), and ultrarapidmetabolizer (UM).

FFPE DNA/RNA Extraction - WaxFree Kits

TrimGen’s WaxFreeTM DNA/RNA Extraction kit is designed for the extraction of genomic DNA and RNA from formalin-fixed, paraffin embedded (FFPE) tissues, as well as fresh or frozen tissues.  The kit uses a unique method to extract nucleic acid from tissue and uses a resin mix to remove PCR inhibitors. The DNA extract from a section of fine needle aspiration (FNA) sample is sufficient to perform 10-20 PCR tests. This high yield feature allows WaxFreeTM kits to successfully extract DNA from a variety of clinical samples.  (see publications)

Pathogen Detection - Enterovirus

TrimGen’s eQ-PCRTM technology is a sensitive detection method for identifying pathogens and genetic mutations. It is based on traditional real-time PCR technology, but utilizes special chemistry.  eQ-PCRTM enhances the fluorescent signals, improves the conventional real-time PCR sensitivity and can detect 1-2 copies of pathogen(s) in a reaction. This proprietary technology provides a fast, accurate and highly sensitive closed-tube assay that eliminates contamination and simplifies work flow.

Enteroviruses (EnV) are positive-sense single -stranded RNA viruses associated with a variety of human and mammalian diseases, the most common cause of aseptic meningitis, especially in infants and the immunocompromised patients. The eQ-PCRTM EnV Detection Kit is a real-time PCR kit used to detect enterovirus present in different type of samples. This detection kit is optimized and validated on Applied Biosystems Real-Time PCR System model 7500.

The diagnosis of EnV meningitis normally requires virus culture and the turnaround times are about 2-5 days. TrimGen’s eQ-PCRTM EnV Detection Kit adopts a one-step RT-PCR method to detect EnV in human tissue samples including CSF in 1 hour. The  eQ-PCRTM technology improves detection sensitivity and the kit can detect as low as 2 copies of viral RNA in a testing tube(analytical study). The design covers over 70 serotype of the EnV family.

Sample Requirement
RNA samples extracted from human tissues, CSF and whole blood. Any commercially available RNA extraction kits are acceptable.
